Manucho's 68th minute winner helped Angola defeat Burkina Faso 2-1 in their AFCON Group B opener in Malabo on Sunday night.
The Real Valladolid ace pounced with a clinical strike from distance after Alain Traore cancelled out Mateus's 48th minute opener for the Black Antelopes.
Earlier on, tournament contenders and group favourites Ivory Coast edged minnows Sudan 1-0, with Didier Drogba heading home the decisive effort.
The first half was a drab affair devoid of chances, with Jonathan Pitroipa and Traore threatening on occasion for Angola, while Moumouni Dagano showed intent going forward for Angola.
The deadlock was broken three minutes into the second period, as Mateus cut in smartly from the right before rattling the net with a rasping low effort.
Ten minutes later, Auxerre star Traore equalised with a superb curling free kick from 26 meters out, with Angolan goalkeeper Carlos Fernandes rooted to the spot.
Manucho then popped up with the telling blow with just over 20 minutes to go, thundering home a left-footed drive on the run after Djakaridja Kone was caught in possession.
The Stallions pushed in the closing stages but were unable to breach a determined Angolan backline.
